Loki has never been a blood relation of Thor's in Norse myth or in Marvel Comics (and only Marvel has him being adopted by Odin). Not that Thor's bloodline has anything to do with Mjolnir's enchantment (as others already mentioned).

On a side note, in Norse myth Laufey was Loki's birth mother, not his father.

I think if it was simply that Thor was turned into a woman there would be less people questioning the motion, still weird as it would be. Also I recall that, and this may have been misinterpretation, Loki is Hel's (Hela in the comics) father and her mother was a wolf. Possibly a misinterpretation, but then again it doesn't sound too crazy for Norse mythology.
